What is the best big effect? I wan't something that screams magician and can be a real reputation maker, and I don't mind spending money for it. How about Peter Marvey's Card Thru Window? Thanks for any answers.

The best big effect is making the Statue of Liberty disappear. :-) Are you looking for something that you can carry with you or something that you need to have set up before hand?

I would say that (limiting effects to what you can buy on this site) the most reputation making effects here would be Mystery Box, Rinkey Dinky, Mind Power Deck, and Dragon Thread. All of these effects require a good amount of practice, but if you put in the time and make the presentation your own then these can be awe inspiring tricks. Aside from that, you can totally blow people away if you have a good knowledge of magic with a totally normal (best if borrowed) deck of cards. There are things that you can do that people will believe absolutely can't be done with normal cards if you work hard enough.

For stage and standup, Rich Morattas Fish bowl production is good. Kids love it, and adults are blown away with it. If you are doing walkaround or impromptu, then invest in card videos. Daryl and Michael Ammar videos are really well done and produced. Although specialty card decks are nice to have and use, you will always want a great number of card tricks which can be performed with no setup, and which can be done with a borrowed deck.
