you are free to do it the way you like. but from my point of view in standing position it is more powerful, because there is less suspicion you might have "cheated/switched" the card in the pocket because everybody can see you don't move your hand

You can perform is seated, or standing without a problem.

I would not perform it seated at all. It provides too much opportunity for the spectator to screw up.
